un-install epsxe and re-install it but this time use this guide to set it up correctly. there are quicker ways to fix something like this but doing this will cut down the likelihood of you running into other setup problems and solve your current issue at the same time.

The SPU plugin you're using with ePSXe is either corrupted or missing :
- confirm that the spupeopsDSound.dll file exists in your plugins folder
- If so, then its corrupted (damaged), just re-download it or select/use another plugin (Eternal SPU plugin /or/ ePSXe SPU core).

Apparently the file is not in the plugins folder. Where can I get it?

Pete's web-site for PSX Plugins

PS : there are other plugins, just use NGEMU.COM and head to PSX stuff, you'll get the full credit.

HAVE FUN.
