It'd be nice if you told us what SPU you are using. The GPU tends not to have anything to do with the sound. Did you check all the necessary boxes to enable sound (the top three options on the main SPU selection screen with sound, CDDA, and XA)?

 Anyway, are you sure THE SPEAKERS ARE ON? And did you clean your ears recently? (Ok, that last one was a joke  ) Seriously though, make sure these three boxes are checked:
