try these in order:
1) try restarting your comp 2) try a different GPU 3) try a different CD plugin 4) try mounting the ISO with Deamon Tools and running it as a CD

Some games just don't work with scph-1001. Try a different one.
But if it's the same game, then I dunno why it stopped working all of a sudden. Since you say you run ISOs, then it's not a CDROM plugin. But just the same, try Mooby's ISO driver.
