The button from keyboard:

1 : "1"
2 : "2"
+ : "P"
- : "M"
Home : "H"
Left : left button
Right : right button
Up : up button
Down : down button

There isn't still interface to config it.
And also A & B buttons are the same on keyboard

It is since gamepad addition, in Rev 1095.
I saw it in source code . But it doesn't make nunchuck activate, it is just for simulate the shake in wario and movement of the pad .

I don't think "S"will work because I tried and didn't do anything and I know that the wiimote is still partly emulate if I am correct.

I want to use 'S' for resident evil umbrella chronicles to switch the weapons but it won't. I am stuck with gun and grenades and knife, that's it. You have the same problem ?

You just need to use the up, down, left or right arrows in the keyboard to change your weapon on Umbrella chronicles

Wait, is there any way to emulate the connection of the nunchuck?

I'm trying to run SMG but I'm being pestered by that "Connect [nunchuck symbol] to [wiimote symbol]" screen.
